This manuscript presents an overview of the recent advances made in confocal Raman microspectroscopy to assess the skin barrier function. This is a nicely written compact manuscript, and I suggest publishing it with the optional minor suggestion: I would not entirely agree with the authors’ statement “The main advantages of these methods are the large scanning area and the ability to visually identify inhomogeneity in the SC thickness. The disadvantages include high costs of the devices and the inability to perform structural analysis of the SC components that provide a barrier function.” For example, OCT has a comparable or lower cost than Raman spectroscopy tool but can directly visualize SC and even assess skin barrier function by study drug diffusivity through outer layers of the skin. Perhaps, this topic deserves better discussion on the paper.
